Ezio Auditore leads the Assassin Brotherhood in Assassin's Creed: Brotherhood.

After the events of Assassin's Creed II, Ezio travels to Rome and works to weaken the Borgia family's control over the city. He rebuilds the local Brotherhood, recruits new members, trains them, and sends them on missions across Europe. This leadership role is a major development from his earlier status as a newly initiated Assassin.

Brotherhood was released in 2010 and continues Ezio's Renaissance storyline. The game introduces a large Rome-based open world, an economic system for restoring districts, and recruitable Assassins who can assist Ezio in combat or undertake assignments. Cesare Borgia is the principal human antagonist, while Rodrigo Borgia remains important to the broader conflict.

Niccolò Machiavelli appears as an adviser and Brotherhood member, but he does not lead the organization in the game. Desmond is the modern-day protagonist, and Altaïr belongs to a much earlier generation.
