Jacob and Evie are twin Assassins who travel to London in 1868, during the Industrial Revolution. They work to weaken Templar control while building their own criminal organization, the Rooks.

The twins have different approaches. Jacob is more impulsive and focuses heavily on street gangs and direct combat, while Evie is more investigative and stealth-oriented. Players can switch between them during much of the campaign, although some missions are assigned to a specific sibling.

Syndicate features historical figures including Queen Victoria, Charles Dickens, and Alexander Graham Bell. It is set later than most mainline entries, placing the Assassin-Templar conflict in Victorian London.
