Jacob and his twin sister Evie leave Crawley and travel to London in 1868, during the Industrial Revolution. Jacob establishes the Rooks, a street gang that helps challenge Templar control over the city. He is impulsive, ambitious, and focused on direct action, while Evie is more methodical and searches for an Isu artifact known as the Piece of Eden.

Syndicate lets players switch between Jacob and Evie for many missions, but their roles are not identical. Jacob specializes in close combat and gang warfare, whereas Evie emphasizes stealth and investigation. Jacob therefore is the twin associated with founding and leading the Rooks, not Evie. Henry Green, whose birth name is Jayadeep Mir, is an Assassin ally and London bureau leader rather than one of the Frye twins. The game’s London setting includes railways, factories, and landmarks such as the Palace of Westminster.
