The game is set in ancient Greece during the fifth century BCE, when Athens and Sparta are fighting the long conflict known as the Peloponnesian War. The campaign begins in 431 BCE, the conventional opening year of the war, and allows the player to travel through regions and islands associated with both rival powers.

Players choose either Kassandra or Alexios as the main character. Kassandra is the canonical protagonist in the series' modern database and promotional continuity, although the game permits either sibling to be selected. The player character becomes involved in family conflicts, political struggles, and the Cult of Kosmos while moving through a large depiction of the Greek world.

The Peloponnesian War historically lasted from 431 to 404 BCE, ending with Sparta's victory over Athens. Assassin's Creed Odyssey uses real figures such as Pericles and Socrates, but its hero, supernatural elements, and many events are fictionalized. The setting is earlier than Origins and later than the Bronze Age period shown in Assassin's Creed games featuring the Isu.
