The bridge crosses the Havel River and connects the Berlin district of Wannsee with Potsdam. During the Cold War, the boundary between West Berlin and East Germany ran across the bridge, making it a carefully controlled crossing point.

Its fame grew from several prisoner and spy exchanges. In 1962, American pilot Francis Gary Powers was exchanged there for Soviet spy Rudolf Abel. In 1985, the bridge hosted a larger exchange involving 23 American agents and four Eastern European agents. In 1986, dissident Anatoly Shcharansky was exchanged for four people held by the Soviet bloc.

Glienicke Bridge is sometimes mistaken for a Berlin Wall crossing inside the city. It was instead a road bridge on the outer edge of West Berlin, and its best-known role was diplomatic prisoner exchange rather than everyday civilian travel.
