Dengvaxia was the first dengue vaccine approved for use in humans.

Dengvaxia, also known as CYD-TDV, was developed by Sanofi Pasteur and first licensed in 2015. It is a live attenuated vaccine based on a yellow-fever-vaccine backbone modified to express dengue-virus proteins.

Dengue has four closely related virus serotypes. That biology makes vaccine development difficult because protection against one serotype does not automatically provide equal protection against the others. Clinical evidence showed that Dengvaxia’s benefits and risks depended strongly on whether a person had previously been infected with dengue.

Because of that difference, some national programs restricted use to people with laboratory evidence of prior infection. Dengvaxia should not be confused with malaria vaccine Mosquirix or human papillomavirus vaccine Gardasil; those vaccines target entirely different pathogens.
