The chemical structure of morphine was first determined in 1925.

British chemist Robert Robinson established morphine’s structural formula in 1925, solving a problem that had challenged chemists for more than a century. Morphine had been isolated from opium in the early 19th century, but identifying its exact arrangement of atoms was far more difficult than simply recognizing its powerful medicinal effects.

Morphine is the principal analgesic alkaloid in opium from the opium poppy, Papaver somniferum. Its complex fused-ring structure includes an aromatic ring, a tertiary amine, ether linkage, and alcohol groups. Robinson’s work clarified the framework, but later research was still needed to settle the molecule’s stereochemistry and fully confirm its three-dimensional arrangement.

The date is often confused with later milestones. Morphine was isolated around 1804 by Friedrich Sertürner, while the first successful total synthesis was achieved in 1952 by Marshall D. Gates Jr. The 1925 date therefore refers specifically to structural determination, not discovery, isolation, or laboratory synthesis. Robinson received the 1947 Nobel Prize in Chemistry, in part for his work on alkaloids and plant-derived compounds.
